本論文應用免標記的導模共振感測器來檢測樣本溶液中Interleukin10 (IL-10) (亦稱為介白素-10)，IL-10在免疫系統中扮演著重要的調節角色。它具有抗發炎和免疫調節的功能。我們先以ELISA去測試，酵素結合免疫吸附分析法，Enzyme-linked immunosorbent assay （ELISA）是一種常用於定量檢測特定蛋白質或其他生物分子存在的方法。其基本原理是利用抗體與目標分子相互結合，然後通過酵素對染料的催化作用來測定目標分子的存在量。由於ELISA分析法僅需少量樣品（約100微升），及此具有高度靈敏度，能夠同時檢測多種抗體。因此，它被廣泛應用於分子生物學、生物化學和醫學研究等領域中。其靈敏度落在1600~12.5 pg/ml左右，但ELISA仍有檢驗時間長、成本較高的問題，因此我們利用導模共振感測器來做檢測，此光學感測技術在分析上實驗時間較短且成本較低，在實驗中，我們最初以PBS緩衝溶液作為基準線，反射光的初始數值為120 uW，加入一抗後，上升到167.9-170.5 uW之間，我們在以 PBS 清洗抗體，數值下降為139-140 uW之間，之後我們加入BSA，做為孔隙填補的材料，加入後數值上升為178-180 uW之間，再以PBS做清洗，此時數值掉到170-173 uW之間，之後再加入2*10-9g /ml抗原，此時數值緩緩上升到175-178 uW之間，再以PBS清洗，此時數值緩緩下跌至176.3uW，因為此數值大於加入抗原前的值170-173 uW，因此可以確定導模共振感測器表面有抗原，證實可測得 2*10-9g/ml的IL-10。.

520 3 $a This paper uses a label-free guided-mode resonance (GMR) sensor to detect Interleukin 10 (IL-10) in sample solutions. IL-10 plays an essential regulatory role in the immune system, with anti-inflammatory and immunomodulatory functions. Initially, Enzyme-linked immunosorbent assay (ELISA) was used to test IL-10. ELISA is a commonly used method to quantitatively detect specific proteins or other biomolecules, using antibodies that bind to target molecules and an enzyme that catalyzes a dye to measure the presence of the target. ELISA requires only a small sample (about 100 microliters) and has high sensitivity, allowing simultaneous detection of multiple antibodies. It is widely used in molecular biology, biochemistry, and medical research, with a sensitivity range of 1600~12.5 pg/ml. However, ELISA has drawbacks, such as long testing times and high costs. Therefore, we utilized a GMR sensor for detection, which offers shorter analysis times and lower costs. In the experiment, we used a PBS buffer as a baseline, with the initial reflected light value at 120 uW. After adding the primary antibody, the value increased to between 167.9~170.5 uW. After washing the antibody with PBS, the value decreased to between 139-140 uW. Then, BSA was added as a blocking material, raising the value to 178-180 uW, followed by another PBS wash, which lowered the value to 170~173 uW. Finally, after adding 2 *10-9 g /ml of antigen, the value gradually increased to 175~178 uW, and another PBS wash decreased it to 176.3 uW. Since this value is greater than the pre-antigen addition value of 170~173 uW, it confirms the presence of antigen on the GMR sensor surface, demonstrating the ability to detect 2*10-9 g/ml of IL-10..
