Lieber EXPERIMENTATOR, liebe EXPERIMENTATORIN, dieses Buch ist eine Orientierungshilfe im verwirrenden Dschungel von Zellen, Medien, Seren, Supplementen und Vorschriften. Im theoretischen Teil bietet es einen Überblick über Grundlagen (z. B. Hayflick-Limit, Seneszenz, Zellzyklus, Zelltod, Krebsentstehung usw.), die für das Verständnis und damit letztlich für die praktische Arbeit mit Zellkulturen wichtig sind. Die 4. Auflage wurde gründlich aktualisiert und um Trendthemen wie das Smarte Labor 4.0, Elektronisches Laborbuch und brandaktuelle Neuentwicklungen beim tissue engineering (organs on a chip) erweitert. Für die Praxis relevante Schwerpunkte wie Steriltechnik, Subkultur, Adhäsion und Detachment, Kryokonservierung und Langzeitlagerung von Zellen sowie zellbiologische Methoden werden verständlich dargestellt. Die Erkennung und Beseitigung von Kontaminationen wird eingehend behandelt. Moderne Techniken wie siRNA und die in-vitro-Differenzierung mesenchymaler Stammzellen wurden in dieser Auflage erstmals um zwei neue Beiträge über den Vergleich verschiedener Isolierungsmethoden extrazellulärer Vesikel und die Kultur von humanen pluripotenten Stammzellen erweitert. In der 4. Auflage findet der Leser neben einer umfassenden Aktualisierung der Ersatzmethoden für Tierversuche auch einen neuen Beitrag über Primärkultur und zusätzlich ein nagelneues Kapitel über 3D-Zellkultur mit anschaulichen Praxisbeispielen. Darüber hinaus enthält das Buch eine Sammlung von Quellen für die Beschaffung von Zelllinien sowie Kontaktadressen für Fortbildungskurse und Dienstleistungsangebote rund um die Zellkultur.
