Accurate assessments of female reproductive development are an important component to the sustainable management of the white shrimp (Penaeus setiferus) fishery in South Carolina (SC). Reproductive development is commonly assessed macroscopically based on ovary color, however, macroscopically distinguishing females that have never spawned from those that have spawned previously can be difficult due to similarities in macroscopic appearances. The detection of prior spawning events has been crucial for estimating reproductive output and informing the timing of commercial season openings. The objectives of this study were to: 1) characterize female white shrimp reproductive development and spawning activity as observed through histology; and 2) compare microscopic observations of spawning activity with macroscopic observations to determine the efficacy of macroscopic assessments. Weekly samples of female white shrimp were collected in the Charleston Harbor, SC from March to June, 2022, and ovarian tissues were processed through histology for microscopic assessments. A detailed framework for distinguishing microscopically observed development stages is provided, along with explanations of oocyte atresia. The robust descriptions of spawning phenology observed microscopically were compared with macroscopic observations to assess agreement between macroscopic and microscopic detection of prior spawns, assuming microscopic approaches are accurate. The finding of an average percent agreement of 49% across the 8 weeks when spawning was taking place and a low of 25% agreement suggests that macroscopic detection of spawning activity based on ovary size and color may be an unreliable tool as currently applied. Analyses were then conducted to determine whether additional macroscopic metrics may increase detection of prior spawn, and while some metrics were statistically different between spawned and non-spawned individuals (e.g., milkiness and melanophore density), no macroscopic metrics definitively differentiated between these groups. Alternative methods should consider broadening the reproductive stages used for assessment.
