The bioethanol industry, primarily reliant on corn fermentation, faced challenges from evolving energy landscapes, leading to the need for innovative solutions. The prospect of diversifying the bioethanol industry by redirecting ethanol (EtOH) production into high-value chemical markets, focusing on acetaldehyde (AcH) as a significant derivative, could be highly beneficial. AcH, a major platform chemical, holds the potential to drive higher profit margins and align with global sustainability efforts. Indeed, as the electrification of passenger transportation continues its aggressive growth, the fate of current EtOH production facilities is in doubt. Still, a redirection to sustainable platform chemicals will provide economic and environmental benefits. This dissertation addresses the challenges in chemocatalytic EtOH oxidation to AcH, emphasizing the limitations of conventional metal-based catalysts and the economic considerations in the industry.In response to these challenges, the study introduced yeast alcohol dehydrogenase (YADH) as an alternative to traditional metal-based chemocatalysts for EtOH oxidation to AcH. The enzyme's specificity and versatility made it a promising catalyst, offering high product selectivity and environmentally benign conditions. The role of various biomass-derived compounds like D-fructose and furfural (FAL) as co-substrates was thoroughly studied. The effects of various reaction parameters on a coupled redox cascade reaction using EtOH and FAL as co-substrates were investigated. Rigorous techno-economic analysis revealed a scenario that could generate a large-scale process with ~40% lower total annual operating cost, and certain challenges, including enzyme stability, were identified. Covalent immobilization of YADH was performed to enhance enzyme stability, longevity, and processability with potential applications in continuous reactions.Further studies into process intensification using various reaction engineering studies like fed-batch and reactive stripping were conducted to explore scale-up aspects of the process. Overall, the obtained results suggest that acetaldehyde and furfuryl alcohol can be produced from ethanol and furfural sustainably, using YADH as the sole biocatalyst. The findings in this dissertation highlight the importance of combining various approaches, including biochemical and biophysical characterization, enzyme immobilization, techno-economic analysis, and process intensification for sustainable process development for valorizing biomass-derived platforms.
