Pseudomonas aeruginosa is commonly associated with ventilator-associated pneumonia, a problem exacerbated during the SARS-CoV-2 pandemic by increased ventilator use. Additionally, P. aeruginosa infections can lead to complications in cystic fibrosis patients, burn victims, immunocompromised patients, organ transplants, and leukemia progression. Previous research in the Boon lab has shown that nitric oxide sensing protein, NosP, mediates nitric oxide-induced biofilm dispersal by inhibiting a kinase, NahK. NahK has been implicated in the GacS Multikinase Network (MKN), the network that regulates biofilm formation in P. aeruginosa and controls most major virulence pathways including quorum sensing. Our goal now is to determine how NahK influences quorum sensing through the GacS MKN, as the individual role of NahK has never been determined.Our data illustrates that a P. aeruginosa strain with a deletion of nahK leads to an upregulation of the main Pseudomonas exotoxin, pyocyanin. Pyocyanin is regulated by several quorum sensing pathways, and we hypothesize that the PQS quorum sensing pathway is the driver of this phenotype through our preliminary qPCR data. We characterized the influence of PQS on the pyocyanin phenotype through virulence phenotypic assays, qPCR, and kinetic fluorescence. In addition, we are also investigating how deletions of some of the other components of the GacS MKN affect pyocyanin production and biofilm formation. We hope to cross-compare these GacS MKN deletions in the same strain in the same study and correlate it to activity of RsmA. We are also exploring how the deletion of nahK also affects the Las quorum sensing system.
