Background: Neurodevelopment and cognition are associated with multiple internal and external factors during sensitive periods of development. Two such factors are sleep and mental health. Sleep is a complex behavior, critical for neurological, somatic, and psychological health. Both factors have been related to the brain's structural and functional state and neuropsychological performance. Additionally, mental health factors, such as anxiety and depression in children and adolescents, are significant problems that impair everyday function and increase the risk of severe mental health disorders in adulthood. Moreover, these factors have been linked to structural and functional changes in the brain and cognitive performance. Therefore, this dissertation is centered on examining the associations between sleep measures (social jetlag and sleep stages duration) and mental health (anxiety and depression) as potential factors that could alter brain structural connectivity and cognitive function in late childhood.Methods: Data from participants enrolled in the Adolescent Brain Cognitive Development (ABCD) Study from September 2016 to February 2020 were included in this dissertation. Exposure variables from the two sections comprised in this dissertation were: (1) actigraphy data used to measure sleep inconsistency and sleep duration and (2) baseline T-scores for anxiety, depression, and anxious/depressed syndrome, calculated with the Child Behavior Checklist tools, were used to measure mental health. Brain structural connectivity measurements and neurocognitive measurements were used as the mediators and outcomes, respectively.Each section included three aims. Aims one and two examined the association between the exposures and brain structural connectivity and cognitive function using linear mixed effect models. Mediator' selection for both studies was conducted through elastic net regressions, as penalization was required to account for multicollinearity and multiple comparisons with the brain structural connectivity measurements. Finally, in aim three, we conducted a mediation analysis with bootstrapping, to explore the mediation effect of brain structural connectivity between the exposures and outcomes.Results: Associations between sleep measures and mental health were found for decreased brain structural connectivity integrity and worse cognitive performance. Particularly, social jetlag was a stronger predictor for worse cognitive function in the three cognitive components than sleep stages duration. Additionally, anxiety and anxious/depressed syndrome were associated with a wider set of cognitive tests outcomes compared to depression. Partial mediations were found for the associations between sleep/mental health measures and cognitive function by the integrity of several white matter tracts, mainly the corticostriate, the inferior/superior frontal cortex, and corticospinal tracts. Additionally, the white matter integrity of several tracts was found to be a partial mediator for the association between mental health and cognitive function, particularly between clinical depression and memory.Conclusions: The overall findings of this dissertation elucidate potential effects that behavioral and mental health factors (such as sleep, anxiety, and depression) have on brain structural connectivity and cognitive function. These behavioral and mental health factors during adolescence could potentially result in long lasting alterations to both structural and functional neural development. As sleep and mental health during childhood are a public health matter that has been traditionally neglected, interventions targeting sleep and mental health, as well as epidemiological surveillance, should be pursued.
