From high performance materials, to commodity plastics, synthetic organic polymers are ubiquitous in the modern world. With modularity in their chemical and physical properties, synthetic polymers can be synthesized for highly specialized applications including those in biological systems. Poly(ethylene glycol) (PEG), a polyether, and poly(ethylene imine) (PEI), a polyimine, are extensively used in a wide range of biological and environmental applications. Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) is one of the most useful characterization techniques of synthetic organic polymers. With optimization of sample preparation, spectral acquisition, and data analysis, MALDI-TOF MS spectra were obtained with sufficient resolution and mass accuracy for the determination of polymer end groups. In combination with other analytical techniques as well as selective functionalizations, MALDI-TOF MS was utilized for the determination of the oxidative degradation products of PEG. MALDI-TOF MS also proved useful when characterizing linear poly(2-ethyl-2-oxazoline) prior to deacylation for the formation of linear poly(ethylene imine). The effect of PEI architecture was then explored in the application of gene delivery. Finally, linear and branched PEI were crosslinked and their performance in copper chelation, antimicrobial activity, and CO2 capture was explored.
