Obesity is a major public health concern with a complex etiology that contributes to chronic disease and poor health. In order to better understand these causes, it is important to study the neural mechanisms that control food intake. Previous research has found that feeding behavior is regulated by specific regions of the hypothalamus in both preclinical studies and rodent models. Somatostatin-expressing neurons in the tuberal nucleus (TN) of the hypothalamus have been identified as critical modulators of feeding in mice. Preliminary data in the Correa Lab used a virally-delivered caspase, a protease that is often used to induce programmed cell death, to ablate the TNSST neurons. We found that ablation of the TNSST neurons decreased food intake in female mice during proestrus when circulating hormones such as estradiol are heightened. Interestingly, there was no effect of ablation in males. These findings raise important questions of the potential sex-based difference in the TNSST neuronal population and its potential response to sex hormones. Therefore, this thesis explored the potential existence of sex-based differences in the number of neurons and the responsiveness to estrogens within the Sst expressing cells of the TN. Although, no sex differences were observed in the quantity of Sst+ cells and Esr1-cre+ cells in the TN, dual immuno-FISH analysis showed approximately 30 percent of Sst expressing cells in both males and females co-express Esr1. These findings indicate that a subset of these neurons may be sensitive to estrogens, providing additional insight into the neural regulation of food intake. Further exploration of the role of estrogen signaling in the TNSST neurons may contribute to a complete understanding of the complex regulation of appetite and its dysregulation in obesity disorders.
