Post-traumatic stress disorder (PTSD) is influenced by both environmental and genetic factors. Various protective factors have been identified for PTSD, which might also influence disorder outcomes. Social support is one such potential protective factor for PTSD risk and severity. As of yet, no studies have explored the interaction between molecular genetic vulnerability to PTSD and social support in PTSD. The current study addressed this gap in the empirical literature by examining the buffering role of social support on the relationship between three polygenic risk scores (PRSs; two PRSs for PTSD and one PRS for Depression) and PTSD symptom severity in a longitudinal sample of 2,042 World Trade Center responders of predominantly European ancestry (mean age at 9/11= 37.57, SD= 8.11, 93.3% male). Participants were recruited via the World Trade Center Health Program, and data were collected across monitoring clinical visits. The average PCL total score across all available visits was used to determine PTSD severity. Two social support variables (family supports and coworker supports) were created by summing reports on available family and coworker support while working at the collapse site. All three PRSs were significantly associated with PTSD severity in this sample (β= 0.068-0.121, ps <0.05). Additionally, moderation analysis was conducted to determine the buffering role of social support on the polygenic risk for PTSD. The presence of coworker support significantly moderated the relationship between PRS for depression and PTSD severity (β= -0.0942, p <0.05), with no other significant interactions. This finding suggests that coworker support can potentially mitigate PTSD severity in those with genetic risk for depression. Present results suggest that the PRS can be used as part of a screening measure to identify and treat responders at greater risk of developing PTSD. Additionally, peer support groups may be a valuable treatment modality for at-risk responders with trauma history.
