The present thesis aims to enhance our understanding of calcium, phosphorus, and magnesium metabolism in dairy cows during the periparturient period through various interventions and manipulations of calcium homeostasis. Additionally, it seeks to elucidate the mechanisms governing calcium regulation during this critical period. The first study demonstrated that calcium chelation and milk stasis during early lactation did not induce the traditional calcium- parathyroid hormone (PTH) negative feedback in dairy cows. The second study compared two different prepartum dietary strategies to improve calcium homeostasis postpartum. The first strategy employed a negative dietary cation-anion difference (DCAD) to induce metabolic acidosis, enhancing tissue sensitivity to PTH and increasing the synthesis of 1,25- dihydroxyvitamin D (1.25(OH)2D3), thereby, increasing Ca mobilization from bone, renal Ca reabsorption, and intestinal Ca absorption. The second strategy involved dietary supplementation with zeolite A, which binds dietary phosphorus, reducing its absorption and subsequently decreasing its blood concentration. This reduction in circulating phosphorus is believed to lower fibroblast growth factor-23 (FGF-23) concentrations, thereby increasing 1a-hydroxylase activity and the conversion of 1,25(OH)2D3, leading to enhanced Ca mobilization from bone and Ca reabsorption from the kidneys. The study concluded that both dietary strategies effectively increased postpartum calcium concentrations, albeit through distinct mechanisms - one via metabolic acidosis and the other through phosphorus deficiency. The third study investigated the effects of prepartum calcium chelation and its impact on postpartum Ca homeostasis. Prepartum infusion with calcium chelator resulted in higher calcium concentrations postpartum. Collectively, this work demonstrates the dynamic nature of calcium homeostasis and highlights the various factors influencing Ca concentrations during the periparturient period. These findings contribute to a deeper understanding of mineral metabolism in dairy cows and provide insights for developing effective management strategies to support animal health and productivity during this challenging period.
