Interspecific somatic cell hybrids were constructed from cell lines resistant to antibiotics inhibitory to mitochondrial function. Individual hybrid cultures were isolated in the presence of these antibiotics. The species of mitochondrial genomes present within each culture were determined by utilizing restriction endonuclease and nucleic acid hybridization techniques.

The transmission of parental resistance(s) to hybrid progeny occurred at a high rate and was accompanied by the retention of the mitochondrial DNA species of the resistant parent(s). Antibiotic-mediated selection for parental resistance markers effectively influenced the observed segregation of parental mitochondrial genome populations within these hybrids.

These experiments have led to the generation of cell hybrids which propagate either or both parental mitochondrial genome population(s) under the supervision of hybrid nuclear machinery. The incorporation of active genetic markers affecting mitochondrial function into hybrids and the utilization of such markers to influence the mitochondrial population within these hybrids establishes an approach to the study of mitochondrial genetics and mitochondrial biogenesis within cultured mammalian cell lines.
