Embryos of the viviparous poeciliid fish, Heterandria formosa undergo a 3900% increase in dry weight during development, indicating substantial maternal-embryonic nutrient transfer. Gestation occurs within the ovarian follicle. During development, a highly vascularized pericardial sac envelops the embryonic head and has been implicated in metabolite exchange. The embryonic surface and follicular epithelium are closely apposed to form a follicular placenta.

For most of development, the entire surface epithelium of embryos is bilaminar and possesses apical microvilli, apical coated pits and vesicles, and vesicles displaying features of lysosomes. However, near the end of gestation, the epidermis differentiates and surface cells on all regions of the embryo, except the pericardial and belly sacs, lose their microvilli. Autoradiography of embryos exposed to ('3)H-glycine demonstrated that those regions of the embryonic surface possessing microvillous cells absorbed glycine. Rates of glycine and glucose absorption by embryos were age dependant with the youngest embryos displaying the greatest weight specific rates of absorption. Ultrastructural studies demonstrated that the macromolecules, iron dextran and ferritin, also were absorbed by surface, absorptive cells.

Follicle cells are characterized by apical microvilli, basal infoldings, basal endocytotic vesicles, and lysosome-like vesicles but lack secretory granules. Iron dextran and ferritn injected into females were absorbed by follicle cells and sequestered in dense-staining vesicles, but they did not enter the follicular fluid. However, electrophoretic analysis of follicular fluid polypeptides indicates that some serum polypeptides are transported across the follicular epithelium.

It is concluded from these studies that embryos are capable of absorbing macromolecules as well as low molecular weight compounds. However, transport of serum macromolecules per se across the follicular epithelium is limited and, therefore, low molecular weight compounds may be the primary nutrient source. It is speculated that follicle cells enhance utilization of serum components for embryonic nutrition by absorbing and degrading serum polypeptides and by transporting the resulting low molecular weight products to the embryo.
