Complete polarimeters and ellipsometers deliver the full Mueller matrix that relates input to output polarization states of light upon transmission or reflection. Two such devices were constructed, one imaging with slow mechanical light modulation, and the other using rapid modulation with a point source detector. Their respective theories and methods of operation are rigorously developed. Interpretation of such measurements in crystals requires a rigorous accounting of the intricate process of refractions, reflections, absorptions, and interferences of photons. Models of such accountings are developed herein for general media and with due regard for partial coherence that arises from the finite spectral and angular resolution of measurement devices. These models were used to establish the foundations on which we obtain complete constitutive relations of anisotropic and optically active crystals. Such measurements have been extremely demanding ever since the discovery of optical activity in alpha-quartz over 200 years ago, but are now becoming routine. Principles of coherence are also central to understanding the polarization of luminescence, as it is an intrinsically incoherent scattering process. Luminescence polarization and the Mueller matrix formalism largely diverged ensuing their common origin in the works of Stokes and Soleillet. In a rediscovery of these beginnings, it is shown how luminescence may be generally described by endowing the differential Mueller matrix formalism or Maxwell's equations with source terms.
