Fibronectin is a high molecular weight glycoprotein found throughout the extracellular matrix in humans. First purified in 1948 as a side product of a fibrinogen isolation, it was noted simply as cold-insoluble globulin. Following its independent discovery in fibroblast research, its abundance throughout the body, and its importance in regulating both disease and materials design, fibronectin has demanded the attention of researchers over the past five decades. With demand for the protein rising alongside its expanding utility, purification methods have been developed, iterated, and optimized to provide high quality material at higher yield and in fewer steps. Herein, the design and application of a modified one-step purification of fibronectin from human plasma is reported. Using gelatin affinity chromatography with a stepwise arginine gradient, the results from this study highlight a method for providing fibronectin from human plasma under non-denaturing conditions in less than one half the time currently used in the state-of-the art purification methods.
