Bone remodeling is an essential, delicately balanced physiological process of coordinated activity of bone cells that remove and deposit new bone tissue in the adult skeleton. Bone remodeling can be influenced by chemical and cellular factors external to the bone. When these factors operate within a homeostatic range, bone removal and formation activities of bone cells are balanced, and the bone remodeling cycle ends without a net change in bone volume or mass during tissue turnover. When the balance is negatively altered to result in a net loss in bone volume or mass, diseases such as osteoporosis occur. Understanding the factors involved in bone responses and their mechanisms of action could ultimately lead to safer and more effective treatments for bone diseases. Bone health is linked to many other systems in the body including the endocrine system, the immune system, and the digestive system. In the past, computational modeling has been used to understand the complexities of bone remodeling. However, few of these models address the connection between bone and other physiological systems. To address this gap, the focus of this dissertation is to develop computational models that connect the bone remodeling process to external influences. The focus is primarily on modeling the interactions between short chain fatty acids produced in the gut and the cells involved in bone remodeling. Important interactions involving the hormone estrogen and the immune T regulatory cells are also discussed. The models developed provided biological insights that were experimentally hard to quantify dynamically and point to the need for further data collection in certain areas to fully unravel the crosstalk between systemic mechanisms and bone responses.
