Overnutrition and obesity are frequently associated with elevated levels of inflammation in both circulation and tissues, as well as an increase of sphingolipids (such as ceramides) in organs that are metabolically active and insulin-sensitive (e.g., skeletal muscle and liver), leading to lipotoxicity, insulin resistance, and eventually type 2 diabetes. Obesity is strongly linked with decreases in skeletal muscle quality and function. Therefore, approaches focused on reducing muscle inflammation and improving muscle function in an overnutrition environment are needed. Dairy protein has been shown to reduce inflammation, increase insulin sensitivity, and increase protein synthesis. Therefore, during my Ph.D. studies, I sought to better understand the mechanism behind these effects in dairy proteins.Since the general population has more easily access to whole protein-dense foods, I also investigated the anabolic effects of Cheddar cheese and bovine milk on markers of skeletal muscle protein synthesis. Cheddar cheese and milk are protein-dense foods and have a high content of the branched-chain amino acid, leucine, which is known to independently upregulate muscle protein synthesis. To determine the acute effects of Cheddar cheese and milk on blood amino acid kinetics and the anabolic effects in skeletal muscle in healthy adults, I conducted a randomized controlled trial.To investigate the potential mechanism behind the anti-inflammatory effects of whey protein, I investigated a whey protein-derived peptide, glycomacropeptide (GMP). This 64-amino acid long peptide makes up ~20-25% of whey protein and has been shown to alter the binding of lipopolysaccharide (LPS) to its receptor TLR4. Both LPS and palmitate are increased due to overnutrition, therefore, during my Ph.D. studies, I conducted a study to better the mechanism of GMP in reducing palmitate-induced skeletal muscle atrophy and inflammation. Additionally, dietary GMP given in a whey protein matrix during a high-fat diet intervention in rodents was investigated to determine the effects of GMP on weight gain, insulin resistance, and sphingolipid accumulation in liver and skeletal muscle.Overall, the culmination of these studies has shed light on the potential anabolic benefits of Cheddar cheese consumption, the therapeutic efficacy of GMP in mitigating inflammation and atrophy, and the impact of dietary GMP on obesity-related outcomes.
